Ok, so I did some more looking into this and unfortunately, it doesn't seem as simple as I once thought.
First off, I indeed did not have iTunes library sharing enabled, so I went into the WHS console and enabled it, and restarted the WHS just to be safe.
I verified that library sharing was working by going into iTunes, and sure enough, the MSS library is now there and I was able to play tracks without any issues.
I also discovered that there was an issue with remote access, even though I had never changed any settings whatsoever. One way or another, I refreshed the connection in the WHS console without even changing any settings, and it reported back that remote access was now working properly.
Just as before, I am able to access the WHS remotely through the web browser and view files, etc, but am still unable to use the media streamer.
The iStream app still kicks back the same error as before.
I'm not quite sure what to try next. My instinct tells me it might be worth using an alternate WHS server, perhaps TZO or another, but this seems to be isolated with the media streamer, as I am able to access it remotely. Ideas anybody?

An update... this does not seem to be directly related to the iStream app. I also cannot stream music or video to my browser on my netbook from a remote connection.
However, I can access the file shares, so it is not a port forwarding/external access issue.
I have a sinking feeling that this is all my fault :) I now vaguely remember disabling iTunes media sharing in the WHS console, which would of course be causing this issue. I will post if that fixes this issue.
